Transfer case? No. But do keep the front and rear axles at the same tire diameter, of course. Probably be safe using E53 sizes.

Suspension? Not really. Think about what the spacer does. All it does is change the effective offset of the wheel. Your spacers will also change the hub diameter but ignore that for a moment. As long as you stay within reason, especially if the final offset remains within a factory wheel parameter for your specific vehicle, there should be no ill effects. If you move to the extreme like the "stanced" crowd, or whatever it's called, then yes, you will adversely affect handling and suspension components.

A spacer and wheel arrangement will be a weaker overall assembly than a wheel alone but the combination works fine. Always use adequate length wheel bolts with ANY wheel spacer.
