55s will most likely be taller but different brands in the same size can be different heights. How much taller depends on how tall the 40s are as compared to the 55s.

Speedometer reading will be less than you are actually traveling assuming the 55s are taller. The amount is the same at any speed. Once you know how much the speedometer is off it is easy to convert on the fly. The height difference will determine if the speedometer is off a little or a lot.

Another consideration is how wide your rims are. There is a small chance that the 55s will not work on you rims.

As far as mileage, since at any given speed your RPMs will be lower, your mileage should improve at cruising speed as long as the transmission does not downshift to maintain that cruising speed.
