Keeping the spare key in the car is a good is a good idea too. If a key is inside the car when the car is remotely locked with another key it will be disabled until the car is opened with the same key it was locked. It is a good idea to check if the key left inside will work if the car is opened with the metal blade. A possible downside would be that if someone breaks in the car and pushes the start button he may find out he can leave with the car easy.

I tried to lock the car with one demote and open it with the physical key from the second remote. The engine started with the remote that I had hidden in the car. I don't have comfort access and because of that my car doesn't start with a push of a button.
On the other hand, on my wife's car that has comfort access, the actual remote has to be very close to the dashboard in order to start the car. If the remote is hidden, say in the trunk, the car doesn't start.
Btw, locking the car with one demote or physical key, it doesn't disable the other key. Its like two people with two different keys are using the same car. If one locks it, it doesn't mean the other one can't open the car unless it has the key that locked the car
